Untersuchung des dynamischen Verhaltens roter Blutkörperchen in mikrofluidischen Systemen

Investigation of the dynamic behavior of red blood cells in microfluidic systems

  • Der Blutkreislauf mit seinem weitverzweigten Kapillarsystem ist ein überlebenswichtiges System im menschlichen Körper. Die Fließeigenschaften des Blutes werden maßgeblich durch die Eigenschaften der roten Blutkörperchen beeinflusst. Ein möglichst genaues Verständnis des Verhaltens roter Blutkörperchen im Kapillarfluss ist daher hilfreich, um physiologische Vorgänge und auch Krankheiten zu verstehen. Rote Blutkörperchen können als sehr elastische Membranhüllen beschrieben werden, die mit einer hämoglobinhaltigen Flüssigkeit gefüllt sind. Insbesondere die Membran der Blutkörperchen, die aus einer Lipiddoppelschicht (Plasmamembran) und einem zweidimensionalen Proteinnetzwerk (Zytoskelett) besteht, bestimmt deren viskoelastische Eigenschaften. In dieser Arbeit wurden rote Blutkörperchen im Fluss durch künstliche Kapillaren untersucht und die beobachteten Phänomene anhand mechanischer sowie molekularer Modelle interpretiert. In dieser Arbeit wurden rote Blutkörperchen im Fluss durch künstliche Kapillaren untersucht und die beobachteten Phänomene anhand mechanischer sowie molekularer Modelle interpretiert. Im Kapillarfluss formen sich rote Blutkörperchen mit zunehmender Geschwindigkeit von ihrer bikonkaven Ruheform in eine pantoffelartige Form und schließlich in eine Fallschirmform um, wodurch sich ihr Flusswiderstand verringert. Für beide Formumwandlungen wurde eine kritische Geschwindigkeit abhängig vom Kapillardurchmesser ermittelt und ein Phasendiagramm erstellt. Um die Rolle molekularer Mechanismen für die elastischen Eigenschaften und die Formumwandlung der Blutkörperchen im Kapillarfluss weiter aufzuklären, wurden die Blutkörperchen chemisch (mit Formaldehyd und Diamid) und auch biochemisch (mit ATP) verändert. Aus der Modifikation mit ATP zeigt sich, wie das Relaxationsverhalten und damit die elastischen Eigenschaften von Blutkörperchen durch Stoffwechselprozesse angepasst und reguliert werden können. Die chemischen Modifikationen durch Formaldehyd und Diamid dienen als Modelle für Krankheiten und schädliche Umwelteinflüsse, welche die Deformierbarkeit und damit die Formumwandlung roter Blutkörperchen im Kapillarfluss beeinflussen. Für die chemische Modifikation mit Diamid wurde gezeigt, dass Blutkörperchen in der Lage sind, diese Schäden in engen Kapillaren zu kompensieren. Es konnte ein molekulares Modell vorgestellt werden, das die beschriebenen Phänomene erklärt. Auf der Basis der hier gewonnen Erkenntnisse lassen sich neue Ansätze für verbesserte, theoretische Modelle, für molekulare und physiologische Prozesse im Zytoskelett und mikrofluidische Systeme für diagnostische Anwendungszwecke gewinnen.show moreshow less
  • The blood circulation with its highly branched capillary system is a vital part of the human body. The rheology of blood is strongly influenced by the properties of red blood cells. A precise knowledge of the behavior or red blood cells in capillary flow is therefore desired to gain better understanding of physiologic processes and diseases. Red blood cells can be described as elastic membranes enclosing a liquid volume consisting mostly of hemoglobin. The membrane of the red blood cells which is composed of a lipid bilayer (the plasma membrane) and a two-dimensional protein network (the cytoskeleton) controls their viscoelastic properties. In this thesis red blood cells flowing through artificial capillaries were investigated and observed phenomena were explained using mechanical and molecular models. With increasing velocity in capillary flow red blood cells reshape from the biconcave resting shape into a parachute-like shape with an intermediate slipper-like shape. In this thesis red blood cells flowing through artificial capillaries were investigated and observed phenomena were explained using mechanical and molecular models. With increasing velocity in capillary flow red blood cells reshape from the biconcave resting shape into a parachute-like shape with an intermediate slipper-like shape. Both dynamic shapes exhibit reduced flow resistance. In order to characterize the shape transitions critical velocities of the shape transitions were defined and a phase diagram was established. To further elucidate the molecular mechanisms controlling the elastic properties and thus the shape transitions, the red blood cells were chemically (with diamide and formaldehyde) and biochemically (with ATP) modified. By modification with ATP it was shown how the relaxation behavior of red blood cells is regulated by metabolic processes. The chemical modifications with diamide and formaldehyde served as model systems for potentially harmful environmental stress and diseases. Both of which influence deformability and shape transition behavior in capillary flow. However, the diamide modified cells are obviously able to compensate the oxidative damage when flowing through narrow capillaries. A molecular model was proposed to explain this phenomenon. The here proposed findings may help to improve theoretical models of cellular and physiological processes as well as for the development of microfluidic systems for diagnostic purposes.show moreshow less
